From looking at the files already used in Quake, the file formats are .TGA accompanied by a .DDS the same as the textures for the static models. Each of the individual interactive elements (buttons, loading bars etc) are seperate files that are put together in the in game GUI editor so the elements can be combined and scripted then the GUI editor saves them as a single .GUI script file that the game uses.
